Functional Characterization of the A414G Loss-of-Function Mutation in HCN4 Associated with Sinus Bradycardia

چکیده

Patients carrying the heterozygous A414G mutation in HCN4 gene, which encodes protein, demonstrate moderate to severe bradycardia of heart. Tetramers subunits compose ion channels sinus node that carry hyperpolarization-activated ‘funny’ current (If), also named ‘pacemaker current’. If plays an essential modulating role pacemaker activity. To assess mechanism by results bradycardia, we first performed voltage clamp measurements on wild-type (WT) and mutant expressed Chinese hamster ovary (CHO) cells. These experiments were at physiological temperature using amphotericin-perforated patch-clamp technique. Next, applied experimentally observed mutation-induced changes CHO cells single human cell model developed Fabbri coworkers. The half-maximal activation V1/2 was 19.9 mV more negative than WT (p < 0.001). In addition, dependence (de)activation time constant showed a −11.9 shift 0.001) compared current. fully-activated density, slope factor curve, reversal potential not significantly affected mutation. computer model, cycle length substantially increased, almost entirely due steady-state activation, this increase prominent under vagal tone. introduction passive atrial load into further reduced beating rate, demonstrating even pronounced interactions between atria. conclusion, identified A414G-induced can explain clinically patients gene

The HCN4 channel mutation D553N associated with bradycardia has a C-linker mediated gating defect.

BACKGROUND/AIMS The D553N mutation located in the C-linker of the cardiac pacemaker channel HCN4 is thought to cause sino-atrial dysfunction via a pronounced dominant-negative trafficking defect. Since HCN4 mutations usually have a minor defect in channel gating, it was our aim to further characterize the disease causing mechanism of D553N. METHODS Fluorescence microscopy, FACS, TEVC and patc...

Point mutation in the HCN4 cardiac ion channel pore affecting synthesis, trafficking, and functional expression is associated with familial asymptomatic sinus bradycardia.

BACKGROUND The hyperpolarization-activated nucleotide-gated channel--HCN4 plays a major role in the diastolic depolarization of sinus atrial node cells. Mutant HCN4 channels have been found to be associated with inherited sinus bradycardia. METHODS AND RESULTS Sixteen members of a family with sinus bradycardia were evaluated. Evaluation included a clinical questionnaire, 12-lead ECGs, Holter ...

the effect of functional/notional approach on the proficiency level of efl learners and its evaluation through functional test

in fact, this study focused on the following questions: 1. is there any difference between the effect of functional/notional approach and the structural approaches to language teaching on the proficiency test of efl learners? 2. can a rather innovative language test referred to as "functional test" ge devised so so to measure the proficiency test of efl learners, and thus be as much reliable an...
